The physico-chemical characteristics of purified arginine kinases from prawn and swimming crab were examined. The molecular weights of prawn and swimming crab enzymes were 40,500 and 40,000, respectively. Amino acid analysis indicated that there were some differences in the contents of proline, glycine, methionine, and lysine. The other amino acid compositions of these enzymes resembled each other.

Both enzymes were stable up to 20°C when they were treated for 10 min at various temperature levels. The enzymes lost their activities at temperatures higher than 25°C. They were more stable at pH 8.0 than pH 7.0. The optimum temperature for the enzyme of prawn was about 42°C and that for swimming crab was about 40°C. The pH optima for the activity of arginine kinase of prawn in the forward and in the reverse reactions were found to be 9.0 and 6.1, respectively. For the swimming crab, the similar optimum pHs at 9.2 in the forward reaction and 5.8 in the reverse reaction were observed. Both enzymes were activated most strongly with Mg2 + and Mn2 + followed by Ca2 +, Co2 +, and Fe2 +. The enzymes were not activated by Sr2 +, Cu2 +, or Zn2 +.

The optimum molar ratio of Mg2 +: ATP in the forward reaction of prawn and swimming crab was found to be 1:1, and the molar ratio of Mg2 + : ADP in the reverse reaction was 4:1 in both cases. Kinetic studies indicated that dissociation constants were rather different. In the prawn, dissociation constants for arginine, ATP, AP, and ADP were 0.19,0.31, 0.67, and 0.29 mM, respectively, but in the swimming crab, they were 0.10, 0.18, 0.22, and 0.11 mM, respectively.  相似文献

筛选茯苓高产胞内多糖和胞内三萜的优良液体发酵出发菌株。采用PDA富集固体平板培养与液体发酵培养测定菌丝体生长速率;采用液体发酵策略分析16种茯苓菌株产胞内多糖与胞内三萜的潜能。实验结果表明菌株生长于固体培养基与种子培养基的生长速率之间没有关联性;降低一级种子培养基初始pH值到4.0时能有效缓解茯苓菌株培养物褐化现象;AS5.137胞内多糖含量最高,达377.60±0.10 mg/g,而DB菌株显示出最高的胞内多糖产量,达1.01±0.13 g/L;Y1菌株胞内三萜含量最高,达83.89±4.28 mg/g,而Jingzhou28菌株胞内三萜产量最高,达136.63±26.66 mg/L。就生产茯苓胞内多糖与胞内三萜而言,AS5.137与DB菌株适合作为液体发酵产胞内多糖的出发菌株;Y1,Jingzhou28,Z(z)与Xingpinzhong菌株均较适合作为液体发酵产胞内三萜的出发菌株。  相似文献

为探讨干燥方式对尾巨桉树皮提取物总三萜含量的影响和其杀螺作用机理,实验以尾巨桉树皮为研究对象,比较鼓风干燥和冷冻干燥两种干燥方式对其树皮乙醇提取物总三萜含量及杀螺活性的影响。结果表明,当乙醇体积分数为60%时,鼓风干燥和冷冻干燥的尾巨桉树皮乙醇提取物中总三萜含量最高,分别为10.81%和13.90%。两种干燥方式得到的尾巨桉树皮乙醇提取物对福寿螺均有较强的毒杀活性,在50mg/L的浓度下处理72h,福寿螺的死亡率分别到达93.1%和100.00%;同时尾巨桉树皮提取物还能显著抑制福寿螺离水上爬,抑制效果优于同浓度下的对照药剂茶皂素(P<0.05)。与空白对照组相比,经鼓风干燥的尾巨桉树皮提取物处理后,福寿螺头足和肝脏的蛋白质含量变化不大(P>0.05);经过冷冻干燥的尾巨桉树皮提取物处理后,福寿螺头足中蛋白质含量显著降低(P<0.05),而肝脏中蛋白质含量无明显变化(P>0.05)。鼓风干燥和冷冻干燥的尾巨桉树皮提取物均可有效抑制福寿螺头足部中LDH、AKP和AST/GOT的活性(P<0.05)以及肝脏中ALT/GPT和AST/GOT的活性(P<0.05)。冷冻干燥的尾巨桉树皮提取物中三萜含量以及杀螺活性均高于鼓风干燥的树皮提取物。  相似文献

Two pentacyclic triterpenes isolated from the bark of henna were identified as 3β, 30-dihydroxylup- 20(29)-ene (hennadiol), and (20S)-3β, 30-dihydroxylupane. The assignment of the C-20 configuration in the latter compound was supported by the analysis of Eu(fod)3-induced 1H NMR chemical shifts in the two C-20 epimers synthesized from lupeol.  相似文献

From the rhizomes of Polypodium formosanum, new triterpenoids of the cycloartane group, (24R)-cyclolaudenol and (24R)-cyclomargenol, were isolated as the corresponding acetates, alcohols and ketones, and their structures were established. A new 12-membered macrolide (1) along with two known C-31 and four known C-32 cycloartane triterpenoids (27) were isolated from the tubers of Bletilla striata. Their structures were determined by spectroscopic analysis, including 1D NMR, 2D NMR, and HRESIMS. The chemotaxonomic significance of these isolates, especially C-31 and C-32 cycloartane triterpenoids, are discussed. To our knowledge, this is the first report on the co-occurrence of the unusual 24-methyl, 24-ethyl, and 24,24-dimethyl cycloartane triterpenoids in the family Orchidaceae.  相似文献

从长白山产蓝萼香茶菜[Ralxlosia Japonica(Burm.f.)Hara var.glaucocalyx(Maxim.)Hara]地上全草中分得13个化合物,根据理化常数和波谱学数据分别鉴定为:木栓酮(1)、β-谷淄醇(2)、乙酰熊果酸(3)、齐墩果酸(4)、熊果酸(5)、蓝萼香茶菜素A(6)、2α-羟基熊果酸(7)、2α,3α-二羟基熊果酸(8)、ent-7β,14α,15三羟基-16-贝壳杉烯-3-酮(9)、山香三萜二烯酸(10)、槲皮素(11)、β-谷甾醇3-O-β-D-吡喃葡萄糖苷(12)和果糖(13)。化合物8为第一次从该植物中分得,化合物10为第一次从该属植物中分得。  相似文献

The phylogeny of the genus Cynanchum s. str. is studied using cpDNA spacers and ITS. Morphological, anatomical and latex triterpenoid data are interpreted in light of the molecular results, and discrepancies are discussed. Vegetative characters are better indicators of relationship than floral characters, especially corona characters. The monophyly of all Malagasy species and, nested within the latter, of all stem-succulent taxa is ascertained and the genera Folotsia, Karimbolea, Platykeleba and Sarcostemma are subsumed under Cynanchum. One African species, C. galgalense, is excluded from Cynanchum.  相似文献

研究了几种药用昆虫对灵芝深层培养过程中细胞生长和三萜产物形成的影响。结果表明,添加不同药用昆虫对灵芝细胞生长和细胞内三萜的形成无显著促进作用(P>0.05),但蜣螂虫粉的添加,能显著促进灵芝胞外三萜的形成(P<0.01)。在添加浓度为5g/L时,胞外三萜的产量从对照的220.2±9.7mg/L提高到304.3±11.8mg/L。结果提示药用昆虫蜣螂虫粉中含有促进灵芝胞外三萜形成的功能因子。  相似文献
